If the Mayor, Glory and Willow start off facing each other I'd say Willow. She could just teleport away and take all the time she needs figuring out how to beat the other two. If Glory had Dawn and the gate was about to open, I'd still say Willow, because when Glory turned to face her she could pretty much teleport dawn away or something.
If you forced the three to stay in a closed arena, then I think Willow would go down pretty quick. She's not invulnerable, and although she can enhance her abilities to sustain damage via a spell, she can't become immortal. Glory is Immortal and invulnerable. In a stand up situation she can't be killed. The only way to really kill her was realized via Ben. So in the end, I'd predict even the mayor would go down. Glory would probably just kick her way out of it's stomache.
