S5 for me because that was the season when Spike really earnt Buffy's respect. They make for great foes during that season and then there is a logical progression to the relationship.
I take Spuffy as covering the whole of Spike's time on the show, because dating back to his debut in School Hard he and Buffy made for a great on screen pairing, maybe more so as enermies. They just bought out the best in each other.
