My favorite laugh out loud Spuffy moment is the scene in "Him" when he has to chase her down and tackle her to get the rocket launcher away from her.

So many scenes in something blue make me laugh like crazy. But this in particular:

  Spike: They're strong, and I can't fight. If they get in, I don't know if I can protect you.
  
  Buffy: You think you have to protect me?
  
  Spike: Oh, not with the Girl-Power bit!
