Do you think Spike was referencing Jenny when he said this?
 
No, I always just thought it was Spike assuming that Giles, given his " Watcher fuddy duddyness" wasn't getting any. Little did he know...

Though while we are on the topic of Jenny refernces, I was reading the Pangs shooting script to check a line and found this:

This is pretty morbid, I don't know how anyone would catch a reference like this without reading the script though.

As for the topic, which episode is the reference from?
 
This is from Bargaining Part 1, 6ABB01. Willow is standing on a high arch and telepathically directing the scoobies to kill vampires with the aid of Spike and the buffybot. A vampire attacks Giles and Spike sets the vampire on fire. The vampire dusts and Spike speaks this part.

I don't think this is a reference to Jenny because Giles never had sex with Jenny. It appears to be a reference to the fact that Giles isn't getting any and hasn't since Olivia in Hush. He had sex with Joyce as mentioned at the end of Earshot but otherwise, nothing was going on.

Thanks Ethan Reigns, ya I don't think this was a reference to Jenny, especially not after all those years.
 
I never saw it as a reference to anything other than the fact that Spike thought Giles was boring and absolutely un-sexy, hence why he wouldn't have ever actually "gotten shagged". LOL
