The most lasting relationships of the Jossverse

I'm pretty sure it's Zoe and Wash. I mean, he even let them get married and
have a kid! though it was after Wash died
. Judging by how those two acted around each other, I'd say they had been married for awhile too. We know they met when Mal lost the war and bought the Serenity and Zoe didn't like Wash at first. So, I would bet they even dated a bit before getting married.

I think that pretty much beats anything human in the BTVS/Angel series.
 
Wait; how long has Olivia been Giles overseas booty call? Is it possible the most casual "couple" is the one with the longest history? Oh the epic lulz there would be if it were true.

I'm thirty minutes from getting off the night shift and I can't stop hearing.

As people have said, Darla/Angelus and Spike/Drusilla are the longest running couples for sheer time. Can't comment on DH or any of the comics, as I've not read them.

Zoe/Wash were probably Firefly's longest running couple, thought as a new relationship begins at the end of Serenity, it's impossible to say how long that would have lasted.

Xander/Anya was a couple for quite a while, and Willow/Tara. Buffy/Angel always seems to flare up again when they're around each other, but I don't know if we can count them as being in a relationship after Season 3, since they're apart from each other the majority of the time, and both have their share of other relationships, or potential ones. Kennedy/Willow, once again, could have gone on for a long time after the show's end (and indeed, it seems to have from what I've heard of the comics).
 
The Master/Darla?

It has been shown that her devotion to him rivaled her relationship with Angelus. And we all know vamps are freaky when it comes to sex.
 
If vampire relationships count, then Darla and Angelus have been together the longest seeing as they were together for 150 years, and Spike and Drusilla come in second.

As for human relationships:

Xander/Anya- 3 years
Willow/Tara- 2 1/2 years
Willow/Oz- 1 1/2 years (they got together at the end of season 2 and broke up in mid-season 4)
Buffy/Angel- About 2 years in total because they were sort of on and off in season 3 and they weren't together after Innocence.
Fred/Gunn- 1 1/2 years
Willow/Kennedy- Almost 2 years.
