What moment defines Buffy as "The Slayer"?

I'm going to out on a limb here and say "The Gift" but for slightly different reasons. Yes, she "sacrificed" her life to save not only her world but all the worlds, as well as her sister but that's not the point. The series was at serious risk of cancellation so Whedon was looking to make a big, final statement. One that not just defined her as the current Slayer but the Slayer that only she could be. As we learned through Kendra in season 2 Slayers traditionally had a much different and much shorter life than Buffy, who refuses to give up who she is at her core. Her last words to Dawn are the perfect sum up of the series and her life. But the ultimate is that final shot of her headstone.

I think of either "Me" in Becoming or Anne when she stops being Anne and is Buffy the Slayer again.
 
Prophecy Girl for me too. For me, it's the first time that she really transcends her human limitations in a full embrace of the calling and responsibilities of the Slayer.
 
Well, to me Buffy really defined herself as Buffy The Slayer and all it entails in S5 Checkpoint.

No fights, no cool action moments, no 'going to her certain death' (other slayers have most likely done all of these and bravo for that) but in Checkpoint Buffy recognizes her place in the scheme of things after facing off against a God (plus great fight with mystery-knights), stands up to the Watcher's Council, tells them her friends are great and that they're part of the slaying unit/package, then orders them to give her the info she needs to do her job (and give Giles his job back) and they do.
Why? Because she's Buffy the goddam vampire Slayer, that's why. Defining moment.
