What would happen If Giles decided not to kill Ben. Would Glory was revenge against Buffy/Scoobies? Or would she try to destroy Humanity?

Dawn would've still opened the portal which would mean that Buffy would have still jumped and killed herself, rather than letting Dawn sacrificing herself.
With this in mind, I'm pretty sure that Glory would hunt down Dawn again and re-open the portal. That's what she had on her mind throughout the whole of Season 5, it was her determination, so I think she would attempt to re-open it. I don't necessarily think that she would try and humanity; at times it seemed that she thought she was above humans, so I don't think she'd get much pleasure from that.
However, saying that, I believe that she'd get revenge on the Scoobies as she'd track down Dawn again. They stopped her first attempt, she definitely doesn't want her second attempt ruined and the fact that they've made her stay in our world for longer than necessary. And she'd have more of an advantage without the Slayer around. Yes, Willow and Tara are powerful witches but I don't think they'd be a match for Glory, nor Spike for that matter, as we saw in Blood Ties. I think Willow and Tara could slow her down but at the end of the day I think Glory would've gotten her revenge as well as getting back to her own dimension.

At the time of the last episode, weren't Glory's powers weakening due to her being on the earthly dimension? If so then I don't know if she'd be strong enough to exact revenge, though she'd definitely try. The opening of the portal was a one time thing so she'd be pretty angry at missing her time slot.

Oh yeah, totally forgot that it was a one-time deal.
If I recall correctly, Glory had been in Buffy's dimension for several hundred years, at least, according to the commander of the Knights of Byzantium, waiting for the proper conditions under which the Key could be used to open the interdimensional portal that would allow her to go home. If that were the case, probably those same conditions wouldn't present themselves again for yet another few hundred years, so Glory would just have to wait around until then, try and retrieve the Key once more, and probably have to contend with Fray before she could attempt her homecoming again. In the meantime, she would probably want to keep as low a profile as possible, just as she had done during her time on earth prior to that. Even as powerful as she was, I doubt that she would want to spend a few centuries in constant warfare with mankind while waiting for the next ride back home.

You always have the best stories. :D
I don't think Glory would every be able to open the portal again. Like people have said, it was a once in a forever opportunity that she'd been waiting for for a good long while. But I do think that barring other complications, Glory would have tried *something.* I'm not creative enough to say just what (this is the reason I'm not Joss Whedon... you know, besides being younger and a girl). However, that would be *if* there were no complications. But we saw how crazy both Glory and Ben were getting. They were merging into each other, and I feel pretty sure that once the chance was missed, that would only escalate. I imagine some weird thing with an exploding head. 😛

So I don't think Glory would have been much threat. I predict that within a few days/weeks, she'd either be catatonic, dead, or completely suppressed by Ben.

Joss would find a way to take her out, he wouldn't have the same Big Bad for 2 seasons

I don't think there is a single show that has a series long big bad, except Lost maybe

Well, the Master was the Big Bad in Season 1, and Angel was the Big Bad in Season 2, and they were both back in pretty much the same capacity in Season 8. Maybe Joss is recycling his bad guys.
Something just occurred to me. If Glory were still around, Buffy and Spike could take her back home in his inter-dimensional ship. Problem solved. Or at least turned back over to the beings who expelled her in the first place.
