Which Characters/Actors Have the Best Chemistry

If you are talking about a single character with the audience... I would say pretty much all of the Fanged Four... Angelus, Spike, Darla and Drusilla. All of the actors were terrific as their evil selves, I was always so engrossed with them. I warmed up to them easily and quickly.
Emma Caulfield and Clare Kramer are very charismatic. I loved Glory and Anya the minute they appeared.
For duos/ groups:
-Anya/ Xander: Emma Caulfield and Nicholas Brendon are great at comedy within the show.
-Fang Gang: Wesley, Fred, Gunn, Cordelia, Angel... all were great.
-Giles and Anya: To me, my dream OTP. The hug in Grave? Quite possibly my favourite hug in television.
-Dawn and Tara/ Buffy or Xander. I think Michelle Tranchenberg is a great actress, and has fantastic chemistry, but especially those three characters. Just watch Real Me.
If more comes, I'll add.

My favourites are the following. I think as a couple Angel and Cordy have a great charisma that's fairly believable. I also think that Buffy and Angel are great together too. Finally I love Buffy and Faith (partly because I think Faith's awesome) when they're friends they're great and when they're enemies they're even better. I love the tension and drama that occurs.

I definitely think SMG and David B. have amazing chemistry. From the moment I got into the show, it was obvious that the two of them shared some amazing electricity in their scenes together.
I also think SMG and Eliza had some incredible chemistry. The moment Faith came into the picture, she added something to the show that it had been missing. No matter if they were playing friends or enemies, both girls played off the other so well that it was a match made in TV heaven.
I also think Eliza has wonderful chemistry with David, as well... I thought their scenes on BtVS were amazing, but it was her appearances on "Angel" that convinced me that if Angel were to ever pair up romantically with Faith, it would be epic. Their portrayal of their characters together made me believe they were truly Angel and Faith, not just actors playing roles. I love them as friends and confidantes, but I also think that their relationship has the potential to grow into something amazing and beautiful.

Thought Willow and Riley had really great chemistry when we first meet him in the library, way better than he had with SMG in that scene.

I have time the third person to mention how awesome the chemistry was between Sarah and Eliza. It really helps that Eliza purposely played up the sexual tension between the two.
Really, take Sarah, Eliza, James, and David. Any combination of those four had quite amazing chemistry together, which is part of the reason why I'm so disappointed that Faith was underutilized in the series.

For me, the actors who have the most amazing chemistry together are SMG and David Boreanaz, I immediately saw their characters, they were not just acting, they were truly the characters and I believed in their love-story. I was kind of sorry/disappointed after the episode "Angel" in season1 not to see them together much again for the rest of the season because it was through this episode and at the end with the kiss, that their chemistry started to hit me. <3 So I was a bit put off by the fact that we had to wait the episode Prophecy to see them together again and even there it didn't have the same impact as in episode "Angel". But then, Joss and writers picked up on their wonderful chemistry and put things right in season 2! it was perfect! the focus was really on them and their scenes were really fantastic!!!
Like many of you, I thought Sarah and Eliza's scenes were pure heaven! again, a great pairing with not only 2 great actresses but also charismatic characters! I immediately liked Faith!!! she has such a strong presence on screen!

Yes, my thought too! Their scenes are good, I'm very fond of their meeting in the library with Buffy who's a bit an outsider and also of the scene when Riley comes to her room to ask information about Buffy!

Why wouldn't you? The two of them are amazing together, with great interactions -- that's what I would call chemistry.
I agree with Sarah and Eliza, dam, no wonder I'm a Fuffy fan, those two have intense chemistry!
One thing I like about the couples is that I think most couples were very well chosen, they each have pretty good chemistry (I'm going to exclude Riley and Buffy, these two just seemed weird).
I'm also gonna say Angelus and Spike! Dam great chemistry there!

Friendships:
Cordy/Angel - I adore this friendship. Charisma and David's chemistry is amazing when the relationship is focused more on their friendship. For some reason they lost all their chemistry when the relationship turned romantic (for me at least, I know I'm in the minority with that statement)
Wes/Gunn - J. August Richards and Alexis had great chemistry, especially in Season 2 when the AtS gang is forced out of the Hotel.
Buffy/Willow/Xander - At least in the early years before everything became fractured within the Scoobies. Alyson and Nick in particular have great chemistry that seemed to work in whatever scenario they were put in.
Spike/Fred - James and Amy worked so well together and the Spike/Fred friendship is one of my favourite Spike-relationships. Their chemistry is cute and light and it brings a real sense of care and trust to the screen.
Romantic Relationships:
Darla/Angel – Julie and David have incredible chemistry, which fills the Dangel relationship with heat, passion, tension and emotion. I love their interactions, in particular because the Dangel relationship has so many levels and the chemistry comes through on every single one.
Buffy/Angel – There’s a reason they’re my OTP. Sarah and David’s chemistry is so strong and so wonderful to watch. It’s why Bangel endures so well; every interaction they have is so chock-full of emotion, heat and love. I always wondered if Sarah and David had something going in real life.
Wes/Lilah – Oh-so-hot. I think Alexis and Stephanie had better chemistry than Alexis had with any of his other love interest (yes, including Fred). This chemistry helped turn what could have simply been a sexual relationship into a multi-layered, rich and engaging ship.
Fred/Gunn – cuter than puppies and kittens and bunnies and fairies, these two were my first AtS ship and it’s all down to the chemistry between J. and Amy. They bring so much love and warmth to the relationship, giving the audience a deep connection and a beautiful ship.

Damn, how on earth did I forget Victor/Sierra? *facepalm* Dichen and Enver have such amazing chemistry that the relationship was able to come through even when they were in Doll state. They're beautiful, in every way possible and their relationship is full of love, warmth and longing.

Agreed although Wes / Gunn were competitors for Fred.
SMG and DB have a palpable chemistry. I think it is especially impressive because it takes on different expressions throughout the series. The energy they have as Buffy/Angel is very different to what is going on when they are Buffy/Angelus. It's very compelling as a viewer to see how the charge between them changes with the demands of the story. Even the actors themselves have often remarked on how well they meshed as actors.
I think SMG and ASH have great chemistry too, as do Alexis and Stephanie Romanov. And you know what, DB and NB had great chemistry. Even when Angel or Xander were just talking about the other there was this extra something to it.

oh yes, you're right! I forgot about these two together! they're awesome!!!!🙂😛
I also forgot about the Darla and Angel's scenes!!! both actors had also "sizzling" chemistry! ^^
and I agree as well about the threesome Buffy/ Willow/ Wander in the first seasons but I remember some scenes in S06 where they are just the three of them and again, the magic was still there between them!
