I know Drusilla and Angel are connected based on the whole Sire thing, but why does she initially collapse crying as if in pain when she senses Angel losing his soul if it’s a “good” thing for their family to have him come back into the fold?

It's a bit since I've seen the episode, but it could be that Dru's connection to Angel, and her psychic sensitivity or whatever you call it means she is actually in pain, because she's feeling the pain that Angel is going through at that point. It may be a happy thing later, but at the time it happens, Dru is literally in pain.

Yes, I'd say it's a combination of the two - the sire/childe connection, and given Dru's abilities explains why she picks up on Angel losing his soul and any pain he felt.
